La microscopie de fluorescence est actuellement la technique la plus couramment utilisée en biologie mais elle reste limitée. Dans ce tutoriel, nous aborderons différentes approches basées soit uniquement sur des concepts optiques, ou en association avec un contrôle photophysique des sondes fluorescentes pour atteindre des résolutions nanométriques.

The direct measurement of fluorescent emittters' decay rate and the simultaneous knowledge of their position is a powerful tool for the study of light-matter interaction at the nanometer scale. We show how this can be experimentally achieved with scanning probe fluorescence microscopy and single-molecule fluorescence lifetime imaging microscopy.
